Acid Cleavable Linker

Acid-cleavable linkers remain stable in the blood circulation system at neutral pH (pH 7.3-7.5). This stability allows ADCs to reach and accumulate in their target cells intact. Once the ADC-antigen complex enters the cell, the weakly acidic conditions of the cell endosome (pH 5.0-6.5) or lysosome (pH 4.5-5.0) trigger the hydrolysis of the pH-sensitive linker, thereby releasing the payload drug.
