PROTAC Linkers

PROTAC Linkers are intermediate linkers that connect the two parts of PROTAC (a small peptide residue used to recognize E3 ubiquitin ligase and a ligand that can be recognized by the target protein). The length of the linker is randomly selected, ranging from 12 carbons to more than 20 carbons. An optimal linker will maximize the interaction between the two parts (target protein and E3 ligase), resulting in efficient ubiquitination and ultimate degradation of the target protein. Common ones include: PEGs, Alkyl-Chain and Alkyl/ether. The insertion of PEG also plays a role in increasing the water solubility of the molecule.
